Properties and Biological Behaviour of Coomassie Blue.

چکیده
The demonstration by Hamilton et al. (1948) and Werko et al. (1949) that the analysis of timeconcentration curves of intravenously injected Evans Blue (T-1824) gave estimates of the cardiac output in man that corresponded closely with those obtained by the direct Fick method stimulated a widespread use of the dye method. However, those properties of Evans Blue that led Dawson et al. (1920) to select it from some 60 dyes as a plasma volume indicator, in particular its affinity for protein and the persistence of tissue coloration, detract from its value in studies of the circulation, especially if repetition of observations is required. Alternative indicators have been widely sought, but none can yet be regarded as wholly satisfactory. In a new attempt to find a solution to the problem the distribution and fate in vivo of some 50 compounds were examined in preliminary animal experiments. As a result the dye, Coomassie Blue, was selected for more detailed examination, the results of which are here reported.

Criticism of the use of Coomassie Brilliant Blue G-250 for the quantitative determination of proteins.
The quantitative determination of proteins in biological fluids, using Coomassie Brilliant Blue G-250, was evaluated. Compared with the biuret method, the Coomassie Blue G-250 method needs a much shorter time for analysis and has a greater sensitivity. The sensitivity of the dye for albumin is significantly greater than for globulins. The standard curves for the biuret method are more linear th...
متن کاملDilution characterstics of Coomasie blue in man.
The azo dye Coomassie Blue is relatively non-toxic, has a rapid disappearance rate, and does not cause any skin discoloration (Taylor and Shillingford, 1959). Because it has these properties the dye can be given repeatedly to man at short intervals of time, a particularly important quality for investigations based upon the Stewart-Hamilton indicator dilution curve. Indeed, the dye has already b...
متن کاملDimethylformamide interferes with Coomassie dye staining of proteins on blue native gel electrophoresis.
Blue native gel electrophoresis (BN-PAGE) is used extensively for characterization of mitochondrial respiratory complexes and uses the binding of Coomassie brilliant blue G-250 to visualize proteins. Oxidative modification of sulfhydryl groups of such proteins can be evaluated by labeling with iodoacetamide conjugated to biotin (BIAM) and detected with streptavidin peroxidase on Western blots f...
متن کاملSlow release of molecules in self-assembling peptide nanofiber scaffold.
Biological hydrogels consisting of self-assembling peptide nanofibers are potentially excellent materials for various controlled molecular release applications. The individual nanofiber consists of ionic self-complementary peptides with 16 amino acids (RADA16, Ac-RADARADARADARADA-CONH(2)) that are characterized by a stable beta-sheet structure and undergo self-assembly into hydrogels containing...
متن کاملLipid, Detergent, and Coomassie Blue G-250 Affect the Migration of Small Membrane Proteins in Blue Native Gels
Blue native gel electrophoresis is a popular method for the determination of the oligomeric state of membrane proteins. Studies using this technique have reported that mitochondrial carriers are dimeric (composed of two ∼32-kDa monomers) and, in some cases, can form physiologically relevant associations with other proteins. Here, we have scrutinized the behavior of the yeast mitochondrial ADP/A...
